Hi David, Thanks for developing DNAWorks and for making it open source! It's a wonderful piece of software and I've had good results with it even when using degenerate codons. Can you think of a way that one could constrain solutions so that gapfixed sequences lie on the sense strand (i.e., forward primers) only? Right now, I just generate multiple solutions and then hand pick the ones that have this property, but it would be better if there was a way to explicitly include that in the optimization. -Ron
